How to Read a Peptide HPLC Chromatogram (Buyer Guide)

An HPLC chromatogram is often the most visual page in a peptide lot file—and the easiest to misread. Procurement teams see a clean main peak and assume the lot is “done.” QC teams see a missing method ID and reject the same PDF. Both are reacting to the same artifact for different jobs.
This guide is for B2B buyers—cosmetic formulators, distributors, lab/industrial procurement, and OEM brand teams—who need to interpret a peptide HPLC trace in the context of a Quality & documentation process and a lot COA. It is not a dosing protocol, not a consumer tutorial, and not a substitute for your own analytical methods.

What an HPLC chromatogram is (in buyer language)
High-performance liquid chromatography (HPLC) separates components in a sample over time under a defined method. The detector records a signal vs. time (or volume). The plot of that signal is the chromatogram.
For peptide B2B release, the chromatogram usually supports:
- Identity context — the main peak appears where the method expects the target peptide (alone, this is not always enough; see MS below).
- Purity / related substances — area of the main peak vs. other peaks under the stated integration rules.
- Lot traceability — when the PDF is tied to a batch/lot ID that matches packing lists and the COA header.
Treat the chromatogram as evidence for a named method on a named lot, not as a marketing graphic.

How to read the plot in five passes
1. Confirm you are looking at the right lot
Match lot ID on the chromatogram (or accompanying COA page) to the packing list and PO. Sample vault files illustrate format; production lots need matching IDs. Peptisova’s diligence UX starts at the COA Vault.
2. Find the main peak and the baseline
A typical peptide purity chromatogram shows one dominant peak and smaller related-substance peaks. Note:
- Is the baseline reasonably stable around the main peak?
- Are there large unresolved shoulders that the PDF still calls “single peak”?
- Does the integration window look intentional or arbitrarily cropped?
You do not need to re-process the raw data to ask these questions—but you do need to ask when the plot and the purity claim feel disconnected.
3. Read purity as method-bound area %
Buyer language should be: “HPLC purity by method X on lot Y,” not “this peptide is 99% pure forever.” Soft catalog bands (for example, typical HPLC ≥98% on COA for some Track A SKUs) are suggested bands—confirm with sales and the lot file before PO.
4. Separate identity from purity
HPLC purity does not automatically prove chemical identity. Many B2B specs add MS confirmation where required. Use Quality and the HPLC/MS checklist when your RFQ needs both.
5. Ask what the small peaks are allowed to be
